    Abstract

    Pulsatile signaling represents a major mode of intercellular communication. It allows for the encoding of information through modulating both the amplitude and the frequency of pulsatile signals. Examples of pulsatile intercellular communication include pulsatile hormone secretion and periodic signals of cyclic AMP (cAMP) in Dictyostelium amoebae. Oscillations in intracellular Calcium provide an additional example of frequency encoding of pulsatile signals within cells. Mathematical models based on experimental observations allow us to uncover the molecular mechanism of frequency encoding of pulsatile signals in intercellular communication. Two examples of frequency encoding will be considered: cAMP pulses in Dictyostelium cells and pulsatile patterns of hormone secretion. In both cases mathematical modeling indicates how frequency encoding is based on reversible receptor desensitization. The modeling approach shows that pulsatile cAMP signaling in Dictyostelium is closely related with pulsatile hormone secretion in higher organisms. In both cases there exists an optimal frequency of pulsatile intercellular communication. These results bear on human physiology as it is now clear that most hormones (growth hormone, insulin, gonadotropin-releasing hormone,…) are secreted in a pulsatile manner. The results support the view that the temporal pattern of a hormone is often as important as its concentration in the blood.
